چکیده

Aim: The aim of the study is to evaluate the efficacy of different decalcifying agent used to decalcify bone. Bone is the strongest part in the human. Two types of bones are seen cancellous bone and compact bone. Demonstration of bone is quite difficult than any other tissues. To obtain satisfactory paraffin or celloidin of bone inorganic calcium must be removed from the organic collagen matrix, cartilage and surrounding tissue. Materials and methods: All the samples collected were from the human mandible fixed in neutral buffered formalin for 24 hrs. Cut into pieces ranging from 2cm in size. The samples were decalcified using 10% formic acid, 5% nitric acid, neutral EDTA, HCL,5%,nitric + EDTA . Results: For 5% nitric acid the initial decalcification started on 9 th day, for HCL on 14 th day , formic acid on 25 th day and 5%nitric acid +EDTA on 22 nd day, neutral EDTA on 45 th day.(graph1) The end point of decalcification for 5% nitric acid was 25days, HCl was 34days, 5 % nitric acid +EDTA was 46 days, 10%formic acid was 47days,for neutral EDTA was 61 days. Conclusion: According to our study among the comparison of the five decalcifying agents formic acid proves to be the best with moderate time for decalcification and with ribboning of sections, good nuclear staining and minimal edema.

Evaluation of Decalcification Techniques for Rat Femurs Using HE and Immunohistochemical Staining

Aim. In routine histopathology, decalcification is an essential step for mineralized tissues. The purpose of this study is to evaluate the effects of different decalcification solutions on the morphological and antigenicity preservation in Sprague Dawley (SD) rat femurs. Materials and Methods. Four different decalcification solutions were employed to remove the mineral substances from rat femur...

Some Practical Considerations on Decalcification of Different Types of Bones

This study was carried out to obtain the best technique for preparation of histological sections of bone with minimal artifacts, preserved tissue and high staining affinity. Different bones from adult female rats were gathered and fixed in 10% neutral buffered formalin (10%NBF) then decalcified in 5 different decalcifying agents. Decalcifiers contained strong acids revealed rapid decalcificatio...

Qualitative histological evaluation of hard and soft tissue components of human permanent teeth using various decalcifying agents - a comparative study.

INTRODUCTION Decalcification is the commonly employed technique in histopathology laboratories as a part of calcified tissue preparation for the microscopic examination. Chemical agents are most commonly used for routine decalcification procedures, though some agents adversely affect the tissue integrity and staining properties. AIM To compare the efficacy of various decalcifying agents by hi...

Macrophages Highly Express Carbonic Anhydrase 2 and Play a Significant Role in Demineralization of the Ectopic Calcification.

Vascular calcification is an important risk factor for cardiovascular disease, and is closely associated with all-cause mortality. Recently, it has been revealed that vascular calcification is not a passive precipitation of circulating minerals, but is a process actively regulated through machinery similar to bone formation. During the bone remodeling, osteoclasts execute the bone resorption by...

Effects of prolonged processing in standard and isotonic trichloroacetic acid (TCA) on cellular preservation in bone marrow trephines.

The effects of a combined fixing and decalcifying solution, trichloroacetic acid, on prepared bone marrow specimens were evaluated to try to improve the speed of trephine biopsy processing in our laboratory. The morphological preservation obtained immediately after decalcification was complete was assessed, and also after prolonged immersion in each of two solutions. Trichloracetic acid was mad...
